Introduction | Alpha 1 Antitrypsin (A1AT, gene name SERPINA1) is a protease inhibitor from the Serpin family. It inhibits a variety of proteases, including trypsin, neutrophil elastase, and chymotrypsin. Like other serpins, A1AT binds to the protease in question and interrupts the standard proteolytic mechanism of the enzyme by covalently bonding to the catalytic amino acid; in this case, a serine residue. This causes permanent inactivation of both the serine protease and the serpin. A1AT deficiency allows neutrophil elastase to degrade proteins in collagen unchecked, leading to emphysema, jaundice, and immune dysregulation. |
