| Biological Activity | Arginase (ARG) is a member of the inducible manganese (Mn2+)-containing enzyme family that catalyse the final step of the urea cycle in the liver, converting l-arginine into l-ornithine and urea. To assess its biological activity, an enzymatic activity assay was performed. Briefly, recombinant human Arginase ARG was serially diluted in deionized water. Samples (40 μL) were added to microplate wells together with 10 μL of substrate mixture containing 100 mM L-arginine, 500 mM glycine, and 1.25 mM MnCl2 (pH 10.5). The reaction mixtures were incubated for 2 h at 37 °C. The enzymatic reaction was then terminated by the addition of 200 μL stop solution containing 2 mM o-phthalaldehyde (OPA), 2 mM N-(1-naphthyl)ethylenediamine (NED), 50 mM boric acid, 1 M sulfuric acid, and 0.03% Brij-35. Plates were covered and incubated for 20 min at room temperature, after which absorbance was measured at 520 nm in endpoint mode. One unit of enzyme activity was defined as the amount of enzyme required to produce 1 μmol of urea per minute. The specific activity of recombinant human ARG was determined to be > 24400 pmol/min/μg, as shown in Figure 3. |

| Reconstitution | To keep the original salt concentration, we recommend reconstituting to the original concentration prior to lyophilization (see Concentration) in ddH2O. If a lower concentration is required, dilute in 20 mM Tris, 150 mM NaCl, pH 8.0. If a higher concentration is required, the product can be reconstituted directly in 20 mM Tris, 150 mM NaCl, pH 8.0, though please note that this will change the overall salt concentration. The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex. |
| Storage | Store lyophilized form at 2-8°C for up to 1 month. For longer periods, store lyophilized or liquid at -80°C. Avoid repeated freeze–thaw cycles. |
| Shelf Life | 12 months. |
