EcoRI is expressed and purified from E. coli that carries the recombinant EcoRI gene. The reaction is conducted for 5-10 minutes at 37 °C, and heat-inactivated at 65 °C for 20 minutes.This enzyme is not sensitive to dam, dcm methylation, but sensitive to mammalian CpG methylation.
