| Biological Activity | A binding ELISA assay was conducted to detect the interaction of recombinant Human MBP and recombinant human CALML3. Briefly, biotin-conjugated MBP was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 μl were then transferred to CALML3-coated microplate wells and incubated for 1 h at 37 °C. Wells were washed 3 times with PBST and incubated for 30 min with Streptavidin-HRP, then aspirated and washed 5 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 μl stop solution was added to the wells and the absorbance was read at 450 nm immediately. The binding activity of recombinanthuman MBP and recombinanthuman CALML3 is shown in the figures. The EC50 for this effect is 7.4 µg/ml. |

If a lower concentration is required, dilute in 20 mM Tris, 150 mM NaCl, pH 8.0.
