| Biological Activity | A binding ELISA assay was conducted to detect the interaction of recombinant Mouse ASPN and recombinant mouse LUM. Briefly, ASPN was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 μl were then transferred to LUM-coated microplate wells and incubated for 2 h at 37 °C. Wells were washed with PBST and incubated for 1 h with anti-ASPN polyclonal antibody, then aspirated and washed 3 times. After incubation with HRP-conjugated secondary antibody, wells were aspirated and washed 3 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 μl stop solution was added to the wells and the absorbance was read at 450 nm immediately. The binding activity of recombinant mouse ASPN and recombinant mouse LUM is shown in the figures. The EC50 for this effect is 2.6 µg/ml. |
