SaII is expressed and purified from E. coli that carries the recombinan SaII gene. The reaction is conducted for 5-10 minutes at 37 °C, and heat-inactivated at 65 °C for 20 minutes. This enzyme is not sentitive to dam, dcm , but sensitive to mammalian CpG methylation.
