Directions for use |
Assay Procedure: - Prepare the Bromocresol Green Working Solution by diluting the 5X concentrated Bromocresol Green Solution with double-distilled water at a ratio of 1:4 (i.e. add 5 ml of 5X concentrated Bromocresol Green Solution to 20 ml of double-distilled water).
- Prepare the Albumin Standard Working Solution (3.48 g/L) by diluting the Albumin Standard Solution (34.8 g/L) with double-distilled water at a ratio of 1:9 (i.e. add 0.3 ml of Albumin Standard Solution to 2.7 ml of double-distilled water).
- Set the sample, standard and blank tubes. We recommend setting each standard and sample in duplicate.
- Add 10 µl of double-distilled water to each blank tube.
- Add 10 µl of Albumin Standard Working Solution to each standard tube.
- Add 10 µl of sample to each sample tube.
- Add 250 µl of Bromocresol Green Working Solution to each tube. Mix thoroughly.
- Allow the tubes to stand at room temperature for 10 minutes, then read and record the absorbance at 628 nm or 630 nm.
- Calculate the albumin concentration as follows:
Albumin Concentration (g/L) = [OD of Sample - OD of Blank] / [OD of Standard - OD of Blank] × Concentration of Albumin Standard Working Solution (3.48 g/L) Standard Curve: - Set the standard tubes and dilute the 34.8 g/L standard to the following concentrations with double-distilled water: 13.92, 8.7, 6.96, 4.35, 3.48, 1.74, 0.87 and 0.435 g/L.
- To a tube, add 10 µl of prepared standard, 10 µl of distilled water, and 250 µl of Bromocresol Green Working Solution. Mix thoroughly
- Allow the tubes to stand at room temperature for 10 minutes, then read and record the absorbance at 628 nm or 630 nm.
