The environment-friendly, Single-Step Dewaxing/Antigen Retrieval Buffer is a 20X concentrated rapid pretreatment reagent for paraffin slides. It replaces the traditional multistep process of dewaxing with only one reagent with comparatively shortened operation time. The steps to process the slides are designed in a simplified manner to reduce variability and enhance stability.

1. Dilute the Dewaxing/Antigen Retrieval Buffer (20X) with DI water to 1X Dewaxing/Antigen Retrieval working solution before use. For example: Take 30 ml Dewaxing/Antigen Retrieval Buffer (pH 9) (20X), dilute with DI water to 600 ml.

2. Boil the 1X Dewaxing/Antigen Retrieval (pH 9) working solution in a glass repair box (using a water bath or microwave). Remove the repair box from the water bath or microwave and immerse the slides into the boiling solution (avoid using a metal slide rack).

3. Place the repair box containing the slides again in a water bath or microwave at medium-low power for 15-30 min to repair the antigen. Do not allow the tissue to get dry during the entire process.

4. Bring the repair box containing slides to RT to let it cool naturally and wash the slides with DI water (avoid breaking up the tissue).
