T4 DNA Ligase is a recombinant enzyme expressed in E. coli from Bacteriophage T4 (cloned gene-30). This 55.3kDa protein catalyzes the formation of a phosphodiester bond between the 5' -phosphate and 3' -hydroxyl termini in duplex DNA or RNA, joining the blunt and cohesive end termini. It also repairs single stranded nicks in duplex DNA, RNA or DNA/RNA hybrids. This product can be used for:
- Cloning of restriction enzyme generated DNA fragments.
- Cloning of PCR products.
- Joining of double-stranded oligonucleotide linkers or adaptors to DNA.
- Site-directed mutagenesis.
- Amplified fragment length polymorphism (AFLP).
- Ligase-mediated RNA detection.
- Nick repair in duplex DNA, RNA or DNA/RNA hybrids.
- Self-circularization of linear DNA.

T4 DNA Ligase can be inhibited by NaCl/KCl at a concentrations greater than 200 mM or by heating at 65°C for 10 min / 70°C for 5 min.Storage Store at 4°C for 2-4 weeks. Store at -20°C for long term storage. For long term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze/thaw cycles. Shelf Life 12 months. Molecular Weight 55.3 kDa Buffer Storage Buffer: 50% glycerol, 20 mM Tris-HCl, pH 7.5, containing 50 mM KCl, 1 mM DTT and 0.1 mM EDTA.
