| Biological Activity | A binding ELISA assay was conducted to detect the interaction of recombinant Human MYC and TBP and TRRAP. Briefly, recombinant human MYC was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 μl were then transferred to TBP-coated and TRRAP microplate wells and incubated for 2 h at 37 °C. Wells were washed with PBST and incubated for 1 h with anti-MYC polyclonal antibody, then aspirated and washed 3 times. After incubation with HRP-conjugated secondary antibody, wells were aspirated and washed 3 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 μl stop solution was added to the wells and the absorbance was read at 450 nm immediately. The binding activity of MYC with TBP and TRRAP is shown in the figures. A dose-dependent effect was observed. |
