Reconstitution | To keep the original salt concentration, we recommend reconstituting to the original concentration prior to lyophilization (see Concentration) in ddH2O. If a lower concentration is required, dilute in PBS, pH 7.4. If a higher concentration is required, the product can be reconstituted directly in PBS, pH 7.4, though please note that this will change the overall salt concentration. The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex. |

Biological Activity | VEGFA is known to induce vascular permeability and growth, promote monocyte/macrophage migration, and inhibit cell apoptosis. To test the effect of VEGFA on cell proliferation of the ECV304 endothelium cell line, cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of VEGFA. After incubation for 72 h, cells were observed using an inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10 µl of CCK-8 solution was added to each well of the plate, which was incubated for 1-4 hours at 37°C. The absorbance was then measured at 450 nm using a microplate reader. Cell proliferation of ECV304 cells after incubation with VEGFA for 72 h observed by inverted microscope is shown in Figure 4 and Figure 5. The dose-effect curve of VEGFA is shown in Figure 6, showing that VEGFA significantly promoted cell proliferation of ECV304 cells. The ED50 for this effect is typically 5.58 to 9.98 ng/ml. |
